Activity of chaperonins – HSP, development of potent and effective immunotherapeutic approaches involving these proteins

A. Asea’s department studies hsp activity aiming to develop effective immunotherapeutic approaches using those proteins. This group have found for the first time that exogenous hsp70 has high cytokine activity. The scientists established that widely used adaptogens can affect plasmatic level of hsp. It was shown in experiments on mice that being regularly used, extracts of Siberian ginseng (Eleutherococcus senticosus), snowdon rose (Rhodiola rosea) and magnolia vine (Schisandra chinensis) lead to increase of the plasmatic level of hsp72. Thus an organism interprets taking adaptogens as a peculiar stress, and the resistance of the organism to actual stress situations increases. The scientists have also evaluated the role of hsp in longevity. They supposed that the level of hsp70 long-livers and their descendants show differ significantly from that level in control individuals.

Numerous researches show that C-Jun N-terminal kinases (JNK) participate in tumor growth and in emergence of neuropathic pain. Prof. P. Gerner’s team studied JNK’s role in tumor growth and emergence of pain in the model of skin cancer. It was confirmed that JNK activation in the tumor mass, the spinal marrow and peripheral nerves contributed to the tumor growth. In this investigation, laboratory animals obtained injection of D-JNKI-1 (peptide, which is a selective inhibitor of JNK). As the result, tumor growth became slower, and reaction of laboratory animals at stimulation became less evident that demonstrated abatement in sensation of pain. Contrary, morphine injection (5 mg/kg), which is often used as analgesic at end-stage cancer, did not inhibit tumor growth. Research data show than inhibitors of JNK, such as D-JNKI-1, may be used to treat cancer and pain syndromes.
